Аннотация: For terminal differentiation of the reproductive organs, the female of the human-pathogenic endoparasite Schistosoma mansoni has to be paired with the male. This stimulation is presumably effected by a chemical ligand. The signal leads to a mitogenic activation of stem-cells of the vitellarium. With this new approach presented here, the receptor for the male stimulus and following signal transduction molecules will be identified and characterized. Finally, this receptor will be isolated and used as a new target for the control of schistosomiasis. Up to now, four new clones containing a tyrosine kinase domain have been isolated by RTPCR. One of them is female specifically expressed and shows high homology to receptor tyrosine kinases (RTK). Hybridization of highly conserved regions of RTKs on male and female mRNA shows two furher transcripts only in females. In addition, with a monospecific anti-phospho-tyrosine antibody a 115kd protein has been detected which is phosphorylated and thus activated only in mature, paired females. Furthermore, it could be demonstrated that the GTPase-activator-protein (GAP) and ras are involved in the male-female-interaction of S. mansoni. Германия, Inst. Genetik., Dusseldorf
