Аннотация: Direct Immunofixation Agglutination (DIFA) is a new screening test system for Toxoplasma-specific IgG antibodies. It is easy to perform and shows high sensitivity and specificity. The sera are directly diluted in wells coated with anti-human IgG antiserum and extracted Toxoplasma antigens (the serum dilution buffer contains dithiothreitol for reduction of IgM antibodies). Formalin-fixed Toxoplasma gondii tachyzoites are added. Specific IgG in the specimen reacts with the fixed antigens and antibodies coated to the wells as well as with the formalin-fixed tachyzoites. This leads to the development of multidimensional immune complexes. The DIFA was tested using 2009 unselected sera from pregnant women in comparison to the dye test (DT) and the IgG-specific IFA. The sensitivity and specificity of the DIFA were both about 99.7%. Even sera with high DT and IFA titers showed no prozone phenomenon in the tested dilution of 1:21.
